1) According to the volume of the sample use :0,5ml tubes up to 30µl 1,5ml tubes up to 50-100µl 2) Make a hole with a needle in the tube used as a column. If a 0,5ml tube is used, make a hole in 1,5ml tube and put the small tube in the other one. 3) Add 2-3 mm of sterile glass beads at the bottom of the tube to avoid sepharose leaking from the tube. 4) Complete the volume of the tube with resuspended sepharose 5) Put the tube in a « collection tube ». Spin at 2000rpm for 3 minutes with an horizontal rotor 6) Add the sample directly on the surface of the sepharose. Put a new Eppendorf tube to collect the flow-through. 7) Spin again at 2000rpm for 3 minutes. The purified DNA sample is in the tube.
