CElixir™ is optimized for analytes that are positively charged or amphoteric substances. Until now, CZE has been very erratic in this range due to the limitations of using bare fused silica as the source of EOF. For this reason, many scientists have developed CZE methods at a pH that is higher than optimal for their sample matrix. CElixir™ offers major advantages, when utilized in our recommended optimization procedures. These result in benefits to you, since you can run your method at low pH (1-7) without the variability of EOF. Your methods can now be component dictated rather than capillary dictated. It is recommended that when developing your method with CElixir™ you scout for your optimal pH. The first step will be to get a peak and determine the EOF using CElixir™. The EOF you determine, even in the basic pH range, will be very close to the EOF in the acidic range. This is an advantage to the practitioner in that it is easy to get a peak in the basic range and not have to worry about pH changing when moving to the acidic range. Start with the CElixir™ Accelerator Solution (B) pH 2.5. CElixir™ Accelerator Solution is used as your run buffer and to solubilize and prepare your samples before injection. If your samples do not go into solution or if they precipitate out at this pH, you should either select another pH or add organic solvent to the sample matrix. See the CElixir™ Operating and Trouble Shooting Manual for guidance on this. Be sure to add a neutral marker to the injection for EOF determination. Follow the instructions in the CElixir™ Operating and Trouble Shooting Manual or the HOW TO USE CElixir™ Easy To Apply Instructions Card to condition your capillary and run your CZE with CElixir™. Once you have a peak (usually a peak is found on the first run) determine if you have good resolution and the EOF from the migration time of the neutral marker. Then try pH CElixir™ with Accelerator Solution (B) pH 4.2, 6.2, 8.2, then 9.2. Determine which two pH’s you have the best resolution between. It is likely that your best performance will be between these two pHs. You can optimize your peak area, shape and resolution by adjusting the pH between these two by following the instructions on page 24 of the CElixir™ Operating and Trouble Shooting Manual on CElixir™ pH Adjustment.
