Bioaxial offers a range of improved imaging solutions integrated to confocal microscope platforms. This flexible offering enables the user to easily upgrade existing confocal microscopes for improved resolution. Based on a unique combination of crystal optics and advanced algorithms, Bioaxial's instruments can measure 2D and 3D fixed and live samples labelled with a broad variety of standard fluorophores.

Technology and products Seeing live biological processes occurring with tiny organelles inside the cell in real time is impossible today. Neither electronic nor optical microscopes can achieve this goal. The most performing optical microscopes are limited to a resolution of 200 nanometers whereas the organelles size is often much smaller. Electronic microscopes can see down to the atomic scale but the samples must be kept in a vacuum and coated with a thin gold layer. Therefore it is not compatible with living cells observation. A new family of technologies, called Super Resolution, appeared on the market lately to overcome the diffraction limit of optical microscopes. Bioaxial has developed a breakthrough Super Resolution technology making possible the observation of living cells at the molecular scale well below the current limit of 200 nanometers. This technology opens new possibilities to allow research scientists to make new discoveries, understand disease mechanisms and develop therapies to fight several unresolved diseases such as cancer, Alzheimer, infections and more. Bioaxial's patented Super Resolution technology is based on the illumination of the sample fluorophores by light spots in which the distribution of the light intensity is controlled and "shaped" by an optical effect called Conical Diffraction. The illumination of the sample with different bright spots combined to a proprietary algorithm calculates the position of the fluorophores with a much higher resolution than the diffraction limit and the performance of the best optical microscopes available on the market.

The Conical Diffraction theory was developed by Sir Michael Berry, a reknowned English physicist. Gabriel Sirat, Bioaxial's Chief Technology Officer, developed a specific branch of Sir Michael Berry's theory of Conical Diffraction which led to the production of subdiffraction light shapes thanks to the interaction of polarized light with biaxial crystals. By recording different fluorescence images created by a sequence of different subdiffraction light shapes, a proprietary algorithm retrieves the position of each of the fluorophores contained in the illuminated spot. Currently the CODIM100 is available as an addon module or supplied together with a Nikon Eclipse Ti and the confocal Nikon C2. CODIM100 will soon be available on other microscope brands and models.

Overcoming the diffraction limit The minimum distance between two light emitters which is possible to observe as separate points is called the Diffraction Limit. In advanced optical microscopes, this limit of 200nm prevents Life Sciences researchers from observing fundamental biological processes occurring at a smaller scale. The Bioaxial sub-diffraction light shapes which are shinned on the biological samples provide additional information on the localization of the fluorescence labels inside the sample. The advanced processing of this information allows Bioaxial to overcome the Diffraction Limit and to distinguish light emitters positioned at distances smaller than 200nm.
Applications All fluorophores currently being used in conventional confcal microscopy can also be used with the BioAxial Super Resolution CODIM100 module. For instance the images below show the difference in resolution obtained on the same sample using the same sample preparation method and fluorophores
